Is this all I need to do for the users in each domain to receive mail or I am missing something?

That would depend on where mail is being sent from.

If your goal is to have this qmail server as a kind of multiplexer that receives mail and redirects it to the "real" servers, you would also need to add those domains to control/rcpthosts.

Do I need to add those domains to control/virtualdomains file?

All domains in the virtualdomains file are considered "local" domains (and any entries in smtproutes are ignored). Is that what you want? If so, then you may as well remove the entries from the smtproutes file.
